丙型肝炎病毒在人端粒酶逆转录酶基因转染细胞中持续存在的研究
Persistence and replication of hepatitis C virus in patient′s cells transfected by human telomerase reverse transcriptase gene
【摘要】 目的:观察人端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)基因转染丙型肝炎患者外周血单个核细胞(peripheral blood mononuclear cells,PBMC)长期存活状态下,细胞内丙肝病毒(HCV)是否仍然存在和复制。方法:将外源性hTERT基因导入4例丙型肝炎患者PBMC,RT-PCR检测其在细胞内的表达,并观察细胞生物学特征。RT-PCR检测细胞内HCV RNA正、负链的表达。结果:转染后细胞寿命延长,增殖旺盛,并保持正常细胞生物学特征。4例患者中,1例患者细胞中连续2个月检测到HCVRNA正链,HCV RNA负链间断出现。结论:HCV可在人端粒酶逆转录酶基因转染后丙型肝炎患者PBMC中长期存在和复制,是一种自然感染状态并长期存活的HCV阳性传代的细胞模型。
【Abstract】 Objective: To study the persistence and replication of hepatitis C virus(HCV) in patient′s peripheral blood mononuclear cells(PBMC) transfected by human telomerase reverse transcriptase(hTERT) gene and cultured in vitro.Methods: Four patients′ PBMC were cultured and transfected with hTERT gene,detected the expression of hTERT mRNA by RT-PCR and analyzed the biological characteristics of the cells.The plus and minus strains of HCV RNA in PBMC were detected by RT-PCR.Results: After transfection,stable expression of hTERT mRNA were detected.The cells had an extended lifespan and proliferative potential.HCV RNA plus strain was detected in one patient’s PBMC continuously for 2 months,and minus strain was detected intermittently.Conclusions: HCV may exist and replicate in patient′s PBMC transfected by human telomerase reverse transcriptase gene and cultured in vitro.A stable and natural cell line with HCV replication in vitro is established.
