镉、铅胁迫对匍匐翦股颖叶片中活性氧的影响

【摘要】 通过2种重金属离子和抗霉素A处理匍匐翦股颖Agrotis stolonifera幼苗叶片,利用2,’7’-二氯荧光黄双乙酸酯溶液对重金属胁迫和抗霉素A处理后叶片中产生的过氧化氢(H2O2)反应形成具荧光物质DCF的特性,用荧光光度分析法分析了叶片中H2O2产生与积累规律。所有观测数据均通过统计分析。结果表明,与对照比较,5.0μmol.L-1的抗霉素A处理后匍匐翦股颖叶片中的DCF荧光值是对照叶片的2.41倍,重金属离子处理后叶片中的DCF荧光值增加了6.63%~73.00%。说明抗霉素A与2种重金属离子均极显著地引起叶肉组织中H2O2含量的增加,重金属胁迫确实会诱发植物细胞内ROS的产生。

【Abstract】 By using fluorescence spectrophotometry method,a systematic study was conducted to determine the effect of cadmium and lead ion stress on reactive oxygen species(ROS) in creeping bent Agrotis stolonifera leaf tissue.All experimental numerical values were analyzed by using the ANOVA of the software SPSS 13.0.The results showed that the(antimycin) A and heavy metal ions stress could significantly effect the opposite contents of hydrogen peroxide(H2O2) in leaf tissue.The antimycin A,a coenzyme Q depressor at electronic pipeline in mitochondria,and heavy metal ions could prevail on enhancing of dichlorofluorescein(DCF) fluorescent value.This means that heavy metal ions indeed prevailed on producing ROS in plant cells.
