藏药桑当降低高尿酸血症物质基础的初步研究

【摘要】 目的:初步研究桑当降低高尿酸血症模型小鼠血清尿酸水平的物质基础。方法:用系统溶剂法(极性由小到大:石油醚-乙酸乙酯-正丁醇-水)提取桑当,制成浸膏,用0.5%CMC-Na配成混悬液,连续7天对小鼠灌胃,选择尿酸酶抑制剂氧嗪钾盐,对小鼠腹腔注射后造成高尿酸血症模型,采用尿酸酶-过氧化物酶偶联法测定小鼠血清尿酸水平;筛选出的有效部位用大孔吸附树脂分离纯化,用乙醇梯度洗脱,洗脱物按上述方法重复药效学实验。结果:乙酸乙酯萃取部位可明显降低血清尿酸水平;40%乙醇洗脱部位可明显降低血清尿酸水平。结论:藏药桑当降低血清高尿酸血症的大部分有效物质存在于40%乙醇洗脱部位。

【Abstract】 Objective:To preliminarily study the substance of sangdang on the treatment of hyperuricaemia.Methods:The experimental drugs were systematically extracted by solutions(petroleum ether?ethyl acetate?n-Butanol?water) from sangdang according to solutions’ polarity.Then the each separated composition was prepared by 0.5% CMC-Na liquor and used to raise the mice for seven days.The animal model of hyperuricaemia was made by injecting K-oxonate liquor.URO-PAP measure was used to examine the blood uric acid level.The effective composition was carried out by elution method.Elutropic materials were used to do the same pharmacodynamic experiment again.Results:The ethyl acetate separating composition can remarkably cut down the blood uric acid level of the hyperuricaemia model mice.The separated composition by Ethanol 40% can also remarkably cut down the blood uric acid level of the hyperuricaemia model mice.Conclusions:The main substance treating hyperuricaemia in sangdang exists in the separated composition which eluted by Ethanol 40%.
