转蜘蛛拖牵丝蛋白基因家蚕蚕丝氨基酸组成及其机械性能
Amino Acid Composition and Tensile Properties of Cocoon Silk from Transgenic Silkworm (Bombyx mori) with Synthetic MaSpI Gene
【摘要】 将以绿色荧光蛋白基因为报告基因、含有人工合成的1.6 kb的蜘蛛拖牵丝蛋白基因及转座子pig-gyBac的转基因载体成功导入减秋无滞育家蚕受精卵,得到转蜘蛛拖牵丝蛋白基因家蚕及绿色荧光茧。对转基因家蚕与对照家蚕丝素蛋白进行了氨基酸组成分析,结果表明转基因蚕茧丝素蛋白甘氨酸和丙氨酸的百分含量分别增加了1.65%和1.80%(平均值);对其生丝的机械性能进行了测试研究,结果表明转基因蚕茧生丝的伸长率降低,断裂强度和初始模量增加,且差异均显著,与理论预期结果吻合。结果表明转基因家蚕蚕丝的机械性能一定程度上得到了提高。
【Abstract】 The piggyBac transposon plasmid containing expressed unit of synthetic MaSpI gene and helper plasmid DNA were introduced into eggs of silkworm using microinjection method.Then transgenic silkworms of three generations were fed successfully.The analysis of amino acid composition and the tests of tensile properties of cocoon silk from transgenic silkworm were performed respectively using an 835-50 Hitachi apparatus of amino acid auto analysis and an YG001A electronic single-fibre tensile instron.The results indicated that the percent content of Gly and Ala in the transgenic green fluorescence cocoon fibroin silk increased with an average of 1.65% and 1.80% respectively as contrasted to that of the control and that the strength and initial modulus of fluorescence cocoon silk fibre also increased conspicuously,and the elongation decreased.All these consistented with the theoretical expectation.
