江浙蝮蛇毒精氨酸酯酶Agkihpin的研究 Ⅱ.对鼻咽癌细胞活力、增殖、迁移和细胞形态的影响

【摘要】 目的:将江浙蝮蛇粗毒中新分离纯化的一种精氨酸酯酶Agkihpin应用于鼻咽癌细胞LXC的体外培养,观察Agkihpin对鼻咽癌细胞活力、增殖、迁移和细胞形态的影响,以期探索治疗鼻咽癌的新方法、新药物。方法:将不同剂量的Agkihpin加入细胞培养液中,用四甲基偶氮唑(MTT)法分析细胞活力,细胞计数法观察和分析细胞增殖和细胞迁移。结果:一定剂量的Agkihpin可抑制LXC的细胞活力、增殖、迁移,并可改变细胞形态和杀伤LXC,且剂量越大抑制、杀伤和细胞形态改变越明显。结论:Agkihpin对鼻咽癌的治疗具有潜在的重要意义,Agkihpin具有作为抗鼻咽癌新药物的开发潜力。

【Abstract】 Objective An arginine esterase from crude venom of Gloydius brevicaudus(Stejneger),Agkihpin,was applied for culture of human nasopharyngeal carcinoma(NPC)cell(LXC)in vitro,so that the effects on cytoactive,multiplication,migratory and morphology of LXC can be observed,and the innovation and new medication for the therapy of nasopharyngeal carcinoma can be explored.Methods Different doses of Agkihpin were added into the culture medium,then the cytoactive was analysed by MTT, and the multiplication and migratory of cell were observed and analysed with taking count of cell.Result Certain dose of Agkihpin could inhibit cytoactive,multiplication and migratory,change the morphology of cell,and kill the cell.The more large the dose was,the more obvious effect of inhibiting,changing and killing would be.Conclusion The effects of Agkihpin on cytoactive,multiplication,migratory and morphology of LXC showed that,Agkihpin,with a potential significance to the therapy of NPC,could be developed to a new medication to resist NPC.
