苦瓜蛋白MAP30基因转化番茄的研究

【摘要】 采用CTAB法提取苦瓜叶片总DNA,经PCR扩增、产物克隆及序列测定,获得序列正确的MAP30基因。构建植物表达载体,转化农杆菌,农杆菌工程菌株侵染番茄子叶,诱导出愈伤组织,并分化出芽和根,抗性植株经分子检测,得到在转录水平上表达MAP30基因的转基因番茄植株。

【Abstract】 After total DNA of Momordica charantia L. was extracted by CTAB method, MAP 30 gene was extracted, cloned and sequenced, and its sequence was amplified by PCR amplification,The expression vector of the MAP 30 gene was constructed, transferred into Agrobacterium tumefasiens which was inoculated into tomato cotyledon. The induced calli were induced to produce shoots and roots. Detection of the rooted plants showed that the MAP 30 gene was expressed on transcript level through molecule identification.
