钾维普对未成熟心肌细胞缺血-再灌注损伤保护作用

【摘要】 目的研究钾维普停搏液对未成熟心肌细胞缺血-再灌注损伤的保护作用。方法以乳鼠未成熟心肌细胞为材料建立缺血-再灌注损伤模型,从心肌细胞搏动频率、细胞生存率、心肌酶学等方面观察钾维普停搏液对心肌细胞缺血-再灌注损伤的保护作用。结果与模型组比较,钾维普组肌酸激酶、丙二醛释放量明显降低,心肌细胞搏动频率、细胞生存率及超氧化物歧化酶活性明显升高。结论钾维普停搏液显著减轻未成熟心肌细胞缺血-再灌注损伤,效果优于高钾液或维拉帕米、普萘洛尔的单独作用。

【Abstract】 Objective To study the protective effect of kalium-verapamil-propranolol(KVP) cardioplegia on cultured immature rat cardiac myocytes injured by ischemia-reperfusion. Methods The ischemia-reperfusion injury model of cultured neonatal rat cardiac myocytes was developed. The spontaneous beating,surviving rate,the content of Creatinkinase,Superoxide Dismutase,Malondialdehyde in supernatant after ischemia-reperfusion were detected. Results Compared with the measurements taken from ischemia-reperfusion model group,the CK and MDA in supernatant decreased sharply; the spontaneous beating,surviving rate of cardiac myocytes and the activity of SOD increased highly. Conclusions The KVP cardioplegia can obviously lessen the injury of ischemia-reperfusion in immature cardiac myocytes. The KVP cardioplegia is superior to hyperkalium cardioplegia,verapamil and propranolol.
