昆布水提物对B-MD-C1(ADR+/+)细胞耐药性的逆转作用研究
MDR reversing effect of thallus laminariae PE on drug resistance cell line B-MD-C1(ADR+/+)
【摘要】 目的研究中药昆布水提物(thalluslami-nariaePE,TLPE)对耐药细胞株B-MD-C1(ADR+/+)耐药性的逆转作用。方法应用MTT法测定TLPE逆转多柔比星对B-MD-C(ADR+/+)的细胞毒作用,应用流式细胞仪(FCM)检测TLPE对多柔比星在细胞内的相对浓度和P-gp蛋白表达水平的影响。结果经TLPE作用后,多柔比星对B-MD-C1(ADR+/+)细胞的抑制率显著增高,P<0·01,明显提高了多柔比星进入并聚集于B-MD-C1(ADR+/+)细胞内的浓度,P<0·01;细胞的P-gp蛋白的表达明显降低,P<0·01;以上作用均与TLPE呈浓度依赖性。结论TLPE可提高多柔比星对耐药细胞的细胞毒作用,这种作用可能与抑制细胞P-gp的表达以及增加细胞内药物浓度有关。
【Abstract】 OBJECTIVE:To study the reversing effect of Chinese drug thallus laminariae PE on B-MD-C1(ADR+/+). METHODS:The reversing effect of TLPE on cytotoxicity of ADM was determined by MTT method. Flow cytometry (FCM) was used to examine P-gp expression and the intracellular concentration of ADM. RESULTS: TLPE inhibited the proliferation of B-MD-C1(ADR+/+)cell in a dose dependent manner (P<0.01). The intracellular concentration of ADM in B-MD-C1(ADR+/+)cell was increased significantly but the P-gp expression was decreased. CONCLUSION:TLPE can enhance cytotoxic effect of ADM on drug-resistance cells. This effect is relate to increased intracellular ADM concentration and inhibite P-gp expression in a concentration-dependent fashion.
