缺氧对肝癌BEL7402细胞周期及放射敏感性的影响

【摘要】 目的:研究肿瘤细胞在缺氧及放射情况下细胞周期的改变及细胞存活的情况。方法:将贴壁培养的肝癌细胞株BEL7402分成对照组、缺氧24h组、缺氧48h组、缺氧加照射组和单纯照射组。应用流式细胞仪检测细胞周期的改变,同时应用成克隆分析法观测细胞存活情况。结果缺氧24h与48h组的肝癌细胞株BEL7402存活分数变化不明显,加照射后的实验组则明显下降而缺氧加放射组与单纯放射组比较细胞存活分数明显升高。肝癌细胞株BEL7402细胞缺氧培养后随着缺氧时间的延长(0~48h)S期细胞逐渐减少,而凋亡细胞逐渐增多;G1期和G2/M期细胞变化不明显;单纯放射组可以看到明显的G2/M期细胞阻滞;而在缺氧加放射组看不到明显的G2/M期细胞阻滞,但凋亡细胞明显增高。结论缺氧(1%氧浓度)对肝癌细胞株BEL7402的存活无明显影响;但能保护肝癌细胞株BEL7402使其免受放射线的损伤;缺氧通过影响射线对G2/M期细胞阻滞降低了肝癌细胞株BEL7402对射线的敏感性。

【Abstract】 OBJECTIVE:To research the cell cycle change and cell surviving of the tumor cell undergone hypoxic and radiated condition.METHODS:The BEL7402 liver tumor cells were divided into the control, hypoxia 24 hours, hypoxia 48 hours, hypoxia plus radiation and radiation alone groups. The cell cycle phase was detected with the cytoflowpy and cell surviving fraction with the colon assay. RESULTS: The liver cancer cell BEL7402 surviving fraction did not change obviously after 24 hours or 48 hours hypoxia, whereas, those which were radiated with hypoxia compared with not increased obviously. As the hypoxic time getting longer (0-48 h), the phase S cells decreased and apoptosis cells increased. Whereas those which were in phase G1 and G2/M did not change remarkably. The G2/M phase block was blocked obviously in radiation alone group but not in hypoxia plus radiation group though the apoptosis cells increased. CONSLUSIONS: Hypoxia (1% oxygen) does not affect the survive of liver cancer cell BEL7402 but can prevent it from the damages of radiation. The hypoxia decrease the radiosensitivity through blocking G2/M phase in the liver cancer BEL7402 cell line.
