乳腺癌细胞DNA分析和转铁蛋白受体表达与化疗药物敏感性的关系

【摘要】 目的:研究乳腺癌细胞DNA周期、转铁蛋白受体(CD71)的表达与化疗药物敏感性的关系。方法:利用流式细胞术(FCM)分析36例乳腺癌细胞DNA周期和CD71+细胞表达率,同时用MTT比色法检测化疗药物敏感性。结果:乳腺癌组织CD71+细胞表达率与5-氟尿嘧啶(5-FU)、甲氨蝶呤(MTX)和环磷酰胺(CTX)相对抑制率存在正相关,r值分别为0·485、0·404和0·496,P值分别为0·003、0·014和0·002,而与多柔比星(ADM)、顺铂(DDP)和丝裂霉素(MMC)相对抑制率无相关关系。正倍体细胞除CTX和MMC的敏感例数少于异倍体细胞外,其余药物均多于异倍体细胞,其中5-FU和MTX明显增多,P值分别为0·04和0·029。结论:乳腺癌细胞CD71的表达率与周期性化疗药物的敏感性具有相关性。当肿瘤细胞相处于增殖状态时,对周期性化疗药物的敏感性增高;肿瘤细胞的异倍体细胞对化疗药物的敏感率低于正倍体细胞。

【Abstract】 OBJECTIVE:To study the relationship between cell cycle , CD_ 71 expression and sensitivity of chemistry drug in breast cancer cells. METHODS: Cell DNA cycle and CD_ 71 expression in 36 patients with breast cancer were detected by the flow cytometry (FCM). Meanwhile, sensitivity of chemistry drug in breast cancer cells was detected by MTT. RESULTS: There were positive correlations between CD_ 71 expression of breast cancer cell and the inhibitor rates of 5-FU, MTX and CTX, r=0.485, 0.404, 0.496 respectively, P=0.003, 0.014, 0002, but there were no correlations between CD_ 71 expression of breast cancer cell and the inhibitor rates of ADM, DDP and MMC. The sensitive cases account of chemistry drug especially 5-FU and MTX in diploid cancer cells were much more than those in aneuploid cancer cell, apart from those in CTX, MMC, P=0.04 and 0.029. CONCLUSIONS: There are positive correlations between expression rate of CD_ 71 of breast cancer cells and the sensitivity of periodicity chemistry drug, and the sensitivity of chemistry drug of aneuploid cancer cells is lower than that of diploid cancer cells.
