蛋白质组学技术及其在胃肠道肿瘤标志物研究中的进展

【摘要】 蛋白质组学以其高通量优势为探寻基因功能的奥秘提供了一种新的思路和研究手段。蛋白质组研究在特定事件或环境下某个细胞或某种组织的基因组表达的全部蛋白质,旨在列出全部蛋白质的细目,弄清楚每一个蛋白质的结构和功能及蛋白质群体内的相互作用,对比疾病和健康状态下它们表达水平的变化。每一种生命运动形式都是特定蛋白质群体在不同时间和空间出现并发挥功能的结果,因而蛋白质组研究是我们理解细胞功能和疾病发生、发展过程的中心环节。随着技术的日新月异,蛋白质组学在寻找肿瘤标志物,阐明肿瘤发生、发展及早期诊断和加快药物开发方面具有重要意义。通过复习文献主要探讨了双向电泳、质谱技术、生物信息学等蛋白质组学相关技术的研究进展,并对蛋白质组学应用于胃肠道肿瘤标志物的研究作了回顾与展望。

【Abstract】 Proteomics, the large-scale analysis of proteins, is an increasingly powerful and indispensable technology in molecular cell biology. Obviously, it will contribute greatly to our understanding of gene function in the post-genomic era. Proteome equals to all proteins that expressed by genome of any cell or tissue in particulars. Its purpose is to list all proteins so as to ensure the structures and functions as well as the interactions between proteins groups, to compare their expressing level changes between the ill status and the health one. Each live form is the resulting of special proteins group emerged and functioned at different time and spaces. In a word, proteomic research is the central part that we understand functions of cells as well as occurrences and developments of diseases. Basing on advances in technology, interest is intensifying in applying proteomics to foster a better understanding of cancer processes, develop new tumor biomarkers for early detection of carcinoma, and accelerate drug research. This article brings into focus on the relevant techniques such as two dimensional gel electrophoresis (2-DE), mass spectrometry (MS) and bioinformatics as well as applications for identifying biomarkers of gastrointestinal neoplasms.
