窄范围pH梯度干胶条可提高胃癌细胞蛋白质组表达谱分辨率
The cutting short pH gradient has higher differentiate rate on proteomic expression patterns of high resolution in gastric adenocarcinoma cells
【摘要】 目的探讨不同pH梯度干胶条在提高胃癌细胞蛋白质表达谱分辨率中的作用,为深入研究胃癌的蛋白质组学奠定基础。方法提取胃低分化腺癌细胞MGC-803的蛋白质,分别采用pH 3~10和pH 4~7不同pH梯度干胶条建立胃低分化腺癌细胞MGC-803蛋白质双向凝胶电泳图谱,与相对应pH区域的蛋白质斑点进行比较。结果成功地构建出两种不同pH梯度MGC-803细胞的双向凝胶电泳图谱,经比较两种图谱蛋白质斑点结果显示,运用窄范围pH 4~7干胶条可更有效分辨出pH 3~10部分区域内聚积的蛋白质斑点。结论采用窄范围pH的干胶条可提高双向电泳图谱分辨率,优化了胃癌细胞蛋白质组学研究的技术体系,可有效获取待测靶蛋白。
【Abstract】 Objective Proteomic expression patterns of high resolution on human gastric adenocarcinoma cells were established,which set a foundation of proteomics analysis of gastric cancer cells.Method Proteins of human low-differentiation gastric adenocarcinoma cells MGC-803 were extracted with several kinds of solutions which have differential solubility,and the two-dimensional polyacrylamide gel electrophoresis(2-DE) map of the proteins extracted was established by using the different pH gradient(IPG)drystrips.Results The 2-DE maps of MGC-803 cells was established successfully,using pH gradient 3~10 and pH 4~7 drystrips,the 2-DE maps of pH 4~7 has higher differentiate rate.Conclusion The result indicates the cutting short pH gradient is an effect utility method improving differentiate rate of 2-DE maps and optimization techniques of proteomics of gastric cancer cells.
