【摘要】 目的探讨血小板活化因子乙酰水解酶(PAF-AH)基因多态性与黎族支气管哮喘之间的关系。方法采用等位基因特异性聚合酶链式反应技术等方法,检测108例黎族支气管哮喘患者及115例健康黎族人PAF-AH基因第9外显子Val279Phe基因多态性和酶活性。结果(1)哮喘组与对照组研究对象血浆PAF-AH突变基因型频率、突变等位基因频率间差别均无显著性意义(P>0.05);(2)哮喘缓解组与对照组研究对象血浆PAF-AH活性间差别无显著性意义(P>0.05),二者均高于哮喘发作组(P<0.001);(3)不论是在对照组,还是在哮喘缓解组,Val/Val基因型和Val/Phe基因型血浆PAF-AH活性间差别均有显著性意义(P<0.01),而两组研究对象在同一基因型中血浆PAF-AH活性间差别均无显著性意义(P>0.05)。结论海南黎族哮喘与PAF-AH基因型无关,但其哮喘所处状态与PAF-AH活性有关。更多还原

【Abstract】 Objective To discuss the relationship between bronchial asthma(BA) and the genotype and activities of platelet-activating factor acetylhydrolase(PAF-AH).Methods 108 patients with asthma and 115 normal controls of national Li were studied.The plasma PAF-AH genotype was detected as representative case with 3 different genotypes(Val/Val,Val/Phe and Phe/Phe) by allele-specific poly merase chain reaction(AS-PCR).The PAF-AH activity in plasma was examined by the changes of substrate assay.Results There were no significant difference in the frequency of genotype mutation and allele mutation in cases and controls.Plasma PAF-AH activities of paroxysm group lower than those of recover group and control group(P<0.001).In either control group or recover group there was significant difference of plasma PAF-AH activity between Val/Val genotype and Val/Phe genotype.In the similar genotype,there was no significant difference of plasma PAF-AH activity between the groups of control and recover.Conclusion There was no association between BA of national Li and the genotype PAF-AH.But there were relationship between the state of BA and the activity of PAF-AH.更多还原
