【摘要】 目的定量检测石蜡包埋的淋巴瘤组织中EB病毒(EBV)的表达。方法对TES水浴DNA提取法进行了改良,从石蜡包埋的淋巴瘤组织中提取高质量的基因组DNA,以实时荧光定量PCR方法检测淋巴瘤组织EBV。结果在60例标本中平均提取DNA量为3.97ng(10μm厚切片2片),86.7%(52/60)标本DNA的纯度(260nm/280nm)≥1.6,其中50%(30/60)淋巴瘤组织EBV呈阳性表达(100～106拷贝/ngDNA)。结论采用改良TES水浴法能获取石蜡包埋组织中高质量基因组DNA,使用该新的实验技术可有效地利用现有的标本资源进行与疾病相关科学的研究。更多还原

【Abstract】 Objective Quantitative analysis of the Epstein-Barr virus in paraffin-embedded lymphoma tissues.Methods Modifications were made to the TES method for DNA extraction.High quality genomic DNA was extracted from paraf-fin-embedded lymphoma tissues,and used for real-time fluorescent quantitative PCR detection of the Epstein-Barr virus.Results The average yield of DNA was3.97ng/case(2pieces of10μm sections),with the purity(260nm/280nm)of86.7%(52/60)samples≥1.6.The Epstein-Barr virus was detected in50%(30/60)paraffin-embedded lymphoma tissues(10 0 ～10 6 copies/ng DNA).Conclusion High quality genomic DNA can be obtained from paraffin-embedded tissues by the modified TES method.Disease-related investigation can be performed more effectively with the currently available samples and improved experimental techniques.更多还原
