毛细管区带电泳模式下氨基酸衍生物的手性拆分

【摘要】 运用荧光试剂1,2-苯并-3,4-二氢咔唑-9-乙基氯甲酸酯(BCEOC)作柱前衍生试剂,基于二元手性选择剂协同作用,以-βCD和SDC作为手性选择剂,最佳配比为1.8∶1.8,缓冲液为60 mM pH=9.3的三(羟甲基)氨基甲烷/磷酸(tris-H3PO4)溶液.在15 min内实现了7种单一氨基酸BCEOC衍生物对映体的手性拆分,分离度均在1.2以上,出峰顺序均为-D型先于L-型.

【Abstract】 A simple and rapid method for the chiral separation of amino acids(AAs) derivatives using 1,2-benzo-3,4-dihydrocarbazole-9-ethyl chloroformate(BCEOC) as the derivative reagent by capillary zone electrophoresis(CZE) has been developed.Based on cooperative effect of binary chiral selectors,β-cyclodextrin(β-CD) and sodium deoxycholate(SDC) were investigated as chiral selectors and the optimum molar ratio of β-CD to SDC was 1.8∶1.8.The running buffer was 60 mM of tri-(hydroxymethyl) aminomethane/phosphate(tris-H3PO4,pH 9.3). In this study,each pair of AAs derivatives was separated within 15 min with a resolution more than 1.20,the D-enantiomers for all tested amino acid derivatives were eluted prior to L-enantiomers.
