紫杉醇对胶质瘤细胞系C6增殖抑制的影响
Effect of paclitaxel on the inhibition of proliferation of glioma cell line C6
【摘要】 目的通过体外实验探讨紫杉醇对胶质瘤细胞系C6的抗增殖作用及其作用机制。方法应用形态学、四甲基偶氮唑蓝实验(MTT)、流式细胞术对紫杉醇诱导的胶质瘤细胞系C6进行检测和观察。结果①紫杉醇1nmol/L剂量组镜下观察细胞形态无明显变化,贴壁状况良好;紫杉醇10、100nmol/L剂量组可见细胞密度减小,少许贴壁生长,偶见坏死细胞;紫杉醇1 000nmol/L剂量组细胞贴壁状况不好, 大部分细胞漂浮于培养液中,部分细胞已坏死;②通过MTT观察到紫杉醇各剂量组与对照组相比抑制率均增高(P<0.05),随着时间的延长,各剂量组不同时间段之间的抑制率增高(P<0.05);紫杉醇对C6的抑制率有剂量和时间的依赖关系;③流式细胞仪分析结果显示细胞有G2-M期阻滞,1 000nmol/L紫杉醇分别作用24、48h后,G2-M期细胞比例分别为63.75%、37.20%(同对照组比较P<0.01)。紫杉醇作用48h时出现凋亡峰,细胞凋亡率为14.31%。结论紫杉醇对胶质瘤细胞系C6具有明显的生长抑制作用,呈时间-剂量依赖性;紫杉醇可以诱导胶质瘤细胞系C6细胞发生凋亡,凋亡与G2-M期阻滞密切相关,G2-M 期阻滞达到一段时间后才诱导细胞凋亡。
【Abstract】 Objective To assess the anti-proliferative effect of paclitaxel on glioma cell line C6 in vitro.Methods The experiment was completed in the experiment center of the First Affiliated Hospital of Jinzhou Medical College from April to July in 2005.Cell morphology, MTT assay and flow cytometry were used to examine paclitaxel- induced apoptosis and growth inhibition effect on glioma cell line C6 in vitro.Results Cells treated with paclitaxel had much changes in morphology:in group of 1nmol/L there had not much changes under an inverted light microscop,cells grew well on the wall of culture flask.After exposure to 10nmol/L,100nmol/L paclitaxel,density of cell was declined,few of cells began to necrose. Most of cells in group of 1000nmol/L detached from culture flask ,necrotic cells became abundant and floated in culture medium;the rates of inhibition in experimental groups were high than those in control groups in MTT assay (P<0.05),the rates of inhibition in every experimental groups of different time quantum became higher with time passing(P<0.05).The growth inhibition effect of paclitaxel on C6 cells was of time- and dosage- dependence; The result of flow cytometry showed that cells were blocked in G2-M phase.after exposure to 1 000nmol/L paclitaxel for 24h and 48h,the proportion of cells in G2-M phase was 63.75% and 37.20%(compared to control group,P<0.01).The apoptosis peak appeared after exposure to paclitaxel for 48h,the cells rate of apoptosis was 14.31%.Conclusion Growth inhibition effects of paclitaxel present on glioma cell line C6,the growth inhibition effect is of time- and dosage-dependence.Paclitaxel can induce apoptosis in C6 cell,apoptosis is correlated nearly to the block in G2-M phase, apoptosis is induced in C6 cell after cells blocked in G2-M phase for some times.
