【摘要】 目的:探讨补肾生血药对缺血后肢大鼠外周循环中骨髓来源内皮祖细胞(EPC)的影响及缺血组织血管新生的影响。方法:建立大鼠后肢缺血模型,喂食补肾生血药,1周后细胞培养观察外周血EPC数量变化,4周后检测缺血组织微血管密度及血管内皮生长因子(VEGF)的表达情况。结果:实验组大鼠外周血培养的EPC(89.1±10.3/mm2)明显高于对照组(48.6±7.5/mm2)(P<0.01),缺血组织微血管密度(288.76±12.54)和VEGF表达(0.233±0.014)均高于对照组(224.84±5.87),(0.162±0.018,P<0.05),且外周血EPC数量与微血管密度之间具有显著相关性(P<0.01)。结论:补肾生血药通过动员骨髓EPC的迁移、分化、促进局部的血管新生,达到改善供血目的。更多还原

【Abstract】 Objective:To explore the effect of limb ischemia on mobilization of bone marrow derived-endothelial pregenitor cells(EPCs) and neovascularization of ischemic tissue with traditional medicine.Methods:Unilateral ischemic model of the hindlimbs was produced by excision of the common and superficial femoral artery in 20 male rats.The other 20 animals served as the control group.At 1 week after operation the changing of the circulating EPCs was observed by mononuclear cells culture.Microvesseldensity,the VEGF expression and angiograph of ischemic limbs was detected at week 4.Results:The number of circulating EPCs in experiment group (89.1±10.3/mm2 VS 48.6±7.5/mm2,P<0.01).Microvessel density of ischemic limbs was significantly higher than that in control group (288.76±12.54/mm2 VS 224.86±5.87/mm2,P<0.01).The VEGF expression in experiment group was significantly higer than that in control group (A:0.233±0.014 VS 0.162±0.0186,P<0.01).The number of circulating EPCs was greatly related to the microvessel density and VEGF expression (P<0.01).Conclusion:Local perfusion of ischemic limb can be augmented by improving neovascularization with EPCs mobilization of bushenshengxue traditional medicine.更多还原
