Syk基因的表达与大肠癌关系的研究
Research on the expression of the Syk gene in human colon Carcinoma
【摘要】 目的:探索Syk基因的表达与大肠癌生成及转移的关系。方法:应用RT-PCR技术检测40例大肠癌组织及癌旁正常组织中Syk基因的表达,通过正常组织与癌组织、有无淋巴结转移、浸润深度、临床分期、病理分级进行比较,观察Syk基因的表达与大肠癌生成及转移的关系。结果:40例大肠组织都有 Syk基因的表达,而40例大肠癌组织中只有13例表达(32.5%)(X2=40.755,P<0.05);28例有淋巴结转移的大肠癌组织,4例Syk基因表达(14.3%),12例无淋巴结转移,有9例表达(75.0%)(X2=14.155,P <0.05)。Syk基因表达与淋巴转移、浸润深度、临床分期有关,与年龄、肿瘤分化等级、大小无关。结论:Syk基因表达缺失与大肠癌生成及转移、浸润深度有关。
【Abstract】 Objective:To evaluate the effects of the Syk gene expression in human colonic carcinoma on tumor growth and metastasis. Methods: The expression of Syk gene in the 40 fresh freezed tissues of colonic carcinoma patlents(tumor tissues,adjacent normal tissues) was detected by RT-PCR technique. Results: All normal colonic tissues were detected the expression of the Syk gene. Compared to normal tissues,27 out of 40 colonic carcinoma did not show any detectable Syk expression. And there were significant difference between the two groups(X2= 40. 755,P<0. 05);only 4 coionic carcinoma tissues in 28 patients with lymph node metastasis had the Syk gene expression.9 colonic carcinoma tissues in 12 patients without lymph node metastasis had the Syk gene expression. The Syk gene expression was negatively correlated to lymph node metastasis(X2=14.155,P<0. 05).Furthermore,the expression of Syk gene was correlated with tumor,lymph-node metastasis ,and infiltration,but no correlation was observed with tumor size differentiation degree, clinical period. Conclusions: Normal colonic tissues have expression of Syk gene. The silencing of Syk gene is correlated to growth and metastasis of colonic carcinoma. The Syk gene expression deletion may play an important role in tumor infiltration and metastasis in colonic cancer.
