【摘要】 目的:研究抑癌基因PTEN在非小细胞肺癌(N SCLC)组织中的表达及其意义。方法:收集78例N SCLC及临床资料。采用免疫组织化学SP法检测PTEN蛋白表达情况。结果:PTEN蛋白表达阳性率在N SCLC中为57.69%(45/78),其中在鳞癌为54.00%(27/50),腺癌为64.29%(18/28)(P>0.05);PTEN蛋白表达阳性率随N SCLC分化程度的降低而降低,在高分化组为76.47%(13/17),中分化组为60.00%(24/40),低分化组为38.10%(8/21),低分化组与高分化组、中分化组之间比较差异均有显著性(P<0.01,P<0.05);在Ⅰ、Ⅱ期者为72.92%(35/48),Ⅲ、Ⅳ期者为33.33%(10/30)(P<0.01);有淋巴结转移的病例为47.92%(23/48),无转移的病例为73.33%(22/30)(P<0.05);PTEN蛋白表达阳性者两年生存率为66.67%,表达阴性者为9.09%,两者之间差异有显著性(P<0.01)。结论:PTEN蛋白表达阳性率的降低与N SCLC的分化程度差、TNM分期高、淋巴结转移及病人的生存期降低相关,可能作为判断预后的指标之一。更多还原

【Abstract】 Objective: To investigate the expression of suppressor gene PTEN(phosphatase and tensin homology deleted on chromosome ten) and its significance in non-small cell lung cancer(NSCLC).Methods: 78 specimens and clinical history of NSCLC were collected.The immunohistochemical staining for PTEN was performed by streptavidin-peroxidase method.Results: [WTBZ]The total positive rate of PTEN protein expression in NSCLC was 57.69%(45/78).There was no significant difference between squamous cell carcinoma(54.00%,27/50) and adenocarcinoma groups(64.29%,18/28)($P$>0.05).The expression rate of the PTEN was 76.47%(13/17) in high differentiation group,60.00%(24/40) in moderate differentiation group,and 38.10%(8/21)in low differentiation group,respectively($P$<0.05,$P$<0.01).The expression rate of the PTEN was 72.92%(35/48) in stage Ⅰ～Ⅱ and 33.33%(10/30)in stage Ⅲ～Ⅳ,respectively($P$<0.01).The PTEN expression rates in NSCLC with and without lymph node metastasis groups were 47.92%(23/48)and 73.33%(22/30),respectively($P$<0.05).Two-years survival rate was 66.67% in positive group and 9.09%in negative group($P$<0.01).Conclusion: The lower expression of PTEN protein in NSCLC was associated with poorer differentiation,higher clinical stage,lymph node metastasis,and shorter survival time.The lost of expression of PTEN may serve as one of markers for poor prognosis.更多还原
