【摘要】 为了探讨新同源盒基因BP 1在乳腺癌的表达,运用生物信息学方法设计19肽,合成并偶联到大分子载体KLH上制成人工免疫原,制备兔抗BP 1多克隆抗体IgG.蛋白印迹方法检测BP 1在乳腺癌细胞系M CF 7、M DA-M B-231细胞均有表达:免疫组织化学结果显示,BP 1蛋白在乳腺癌的表达率显著高于癌旁组织,在雌激素受体阴性肿瘤的表达率高于雌激素受体阳性组。BP 1基因的异常表达参与了乳腺癌的发生,是一种新的乳腺癌分子标志物.更多还原

【Abstract】 The present study is to explore the expression of BP1,a novel homeobox gene,in breast cancer.The specific oligopeptide(19AAs) was selected by bioinformatics,synthesized and conjugated to a high molecular carrier KLH.A polyclonal rabbit anti-BP1 antibody IgG was prepared.Expression of BP1 was detected positively in MCF-7 and MDA-MB-231 breast cancer cell lines by western blot.Then,anti-BP1 IgG was used to determine the expression BP1 in 28 pairs of breast cancer and noncancerous breast tissues by immunohistochemistry.The positive rate of BP1 expression in breast cancer was markedly higher than that in noncancerous tissue.There was significant difference in BP1 expression between ER(estrogen receptor)-negative and ER-positive tumors.BP1 gene is a gene closely related to breast carcinogenesis,it may be a novel molecular marker for breast cancer.更多还原
