嗜热解烃菌NG80-2的鉴定及其特性
Identification and Characters of Thermophilic Desmolysing Bacteria NG80-2
【摘要】 利用细胞形态、生理生化特征、不同碳源发酵产酸试验及16S rDNA的进化树分析的方法,对分离获得的嗜热解烃细菌NG 80-2进行了鉴定,确定为G eobacillus therm od en itrif icans,其16S rDNA与G eobacillustherm od en itrif icans BG SC 94A l的相似性最高,达到99.80%.该菌最适生长温度为65℃,最适pH为7.2,能在以原油为唯一碳源的无机盐培养基中生长,并且能选择性降解C 15~C 36直链烷烃,其中对正-十六烷(C 16)的降解率高达58.9%.优良的烃降解性能对微生物开采重质原油和治理石油污染具有潜在的应用价值.
【Abstract】 A thermophilic desmolysing bacteria NG80-2,identified as Geobacillus thermodenitrificans according to morphological,physio-biochemical characters and use of different carbonhydrate experiments was studied.16S rDNA sequence evolutionary analysis showed that NG80-2 has similarities of 99.80 % to Geobacillus thermodenitrificans BGSC 94A1.The optimum temperature and optimum pH of NG80-2 is 65 ℃ and 7.2,respectively.This strain also grows well on the inorganic salt medium with crude oil as sole carbon source,and can selectively degrade C15~C36 n-alkane,among of which degradation rate of n-hexadecane was up to(58.9 %).According to the good characters of alkane-degradation,NG80-2 may be potentially used for oily-waste treatment.
