知母药材苷元类成分RP-HPLC-ELSD指纹图谱研究

【摘要】 目的:采用RP-HPLC-ELSD法建立知母药材的指纹图谱。方法:以Krom asil C18(250.0 mm×4.6 mm,5μm)为色谱柱,采用甲醇-水进行梯度洗脱,流速1.0 m l.m in-1,柱温30℃,检测器温度100℃,气体流速2.0 m l.m in-1。结果:共标出知母药材苷元类成分8个共有峰,重复性、精密度和溶液稳定性试验中,峰面积大于10%总蜂面积的共有峰,其相对峰面积的RSD值均小于3%,符合《中药注射剂指纹图谱研究的技术要求(暂行)》中的有关规定。结论:RP-HPLC-ELSD法稳定、可靠、重复性好,为知母药材质量控制提供参考。

【Abstract】 Objective To establish the fingerprint chromatogram of crude Anemarrhena asphodeloides.Methods HPLC with Kromasil C18(250.0×4.6mm,5μm) column was used,the CH3OHH2O(gradient elution) as a mobile phase and with the column temperature of 30℃.The ELSD detector temperature was 100℃ and the gas flowrate was 2.0ml·min-1.Results 8 peaks were separated respectively on the HPLC fingerprint.The RSD of precision,reproducibility and stability of the solution were less than 3%.Conclusion The method is simple and accurate with a good reproducibility and can be used for a quality control method for Anemarrhena asphodeloides.
