高分子材料对尼莫地平/羟丙基-β-环糊精超分子体系溶解与溶出的影响
Influence of Polymers on the Solubility and Dissolution of Nimodipine/HP-β-CD Supermolecule Systems
【摘要】 应用相溶解度法比较了高分子(PVP、PEG、HPMC)对HP-β-CD体系稳定性和药物溶解性的影响.采用冷冻干燥和物理混合法,制备了药物/羟丙基-β-环糊精二元体系及三元高分子固体体系,测定了各体系的溶出曲线,采用差示热分析考察了各体系的热性质.研究了常见高分子药用辅料对尼莫地平/羟丙基-β-环糊精包合和溶解性的影响.结果表明,三元体系表观稳定常数KS提高后尼莫地平溶解度较两元体系上升,但溶出速率均有所降低.可见,高分子的加入可以改善HP-β-CD增溶尼莫地平的效果,高分子对尼莫地平/羟丙基-β-环糊精体系的溶出速率的影响较为复杂,与高分子种类及体系中的含量有关.
【Abstract】 The aim is to investigate the influence of polymers on Nimodipine / HP-β-CD binary system .Methods:Compare the effect of different polymers(PVP/PEG/HPMC) on the stability and solubility with phase-solubility method. Prepare nimodipine/ HP-β-CD binary system and nimodipine / HP-β-CD / polymers ternary systems with lyophilization and physical mixed methods. These systems are characterized by thermal analysis (DSC) and dissolution studies. Results:The Ks was increased when the ternary system was formulated .Compare the binary system, the solubility is increased and the dissolution rate is decreased .Conclusion: The addition of polymers can improve the solubilization effect of cyclodextrin. The breed and concentration of polymers can affect the dissolution with a complicated mechanism.
