人肺癌和乳腺癌组织IL-15的表达与临床病理因素相关分析

【摘要】 目的检测人体肺癌和乳腺癌组织中IL-15和IL-15Rα的表达及NK细胞和CD8+细胞的浸润,以探求IL-15在人体肺癌和乳腺癌发生发展中的作用。方法用免疫组化方法,检测29例肺癌和51例乳腺癌肿瘤组织标本中IL-15和IL-15Rα的表达及NK细胞和CD8+细胞的浸润,分析IL-15、IL-15Rα、NK细胞、CD8+细胞之间相关关系及与临床病理因素之间的关系。结果肺癌和乳腺癌肿瘤组织中IL-15表达的阳性染色密度值(PSD)随临床分期增加而减少,肿瘤直径<3cm组大于肿瘤直径≥3cm组,无淋巴结转移组大于有淋巴结转移组。两种肿瘤组织中IL-15的表达和IL-15Rα的表达没有相关性,与NK细胞、CD8+细胞的浸润成正相关。IL-15Rα的表达与这两种肿瘤病人的临床分期、肿瘤大小、淋巴结转移无关。结论IL-15高表达者,多处于肿瘤分期早期,瘤体体积较小,少伴有淋巴结转移;IL-15低表达者,多处于肿瘤分期晚期,瘤体体积较大,多伴有淋巴结转移。推测IL-15在这两种肿瘤发生发展中可能起抑制作用。

【Abstract】 Objective To clarify the effects of IL-15 on the development of human lung cancer and breast cancer by detecting the IL-15, IL-15R α, NK cells, and CD8+ cells in the cancer tissues. Methods IL-15, IL-15 Rα, NK cells, and CD8+ cells were evaluated by immunohistochemical staining in 29 cases of lung cancer and 51 cases of breast cancer, for analyzing the relationships between the IL-15, IL-15 Rα, NK cells, CD8+ cells and the clinical characters of human lung cancer and breast cancer. Results In both lung and breast cancer cases, positive staining density (PSD) of IL-15 expression was significantly higher in early stage than in late stage, significantly higher in tumor size<3 cm group than in tumor size≥3 cm group, significantly higher in lymph node un-metastasis group than in lymph node metastasis group, and had no relationship with IL-15Rα expression, but had positive relationship with NK cells and CD8+ cells infiltrating in tumor tissue. In both tumors, the PSD of IL-15Rα expression did not show remarkable correlation with clinical stages, tumor size, and lymph node metastasis. Conclusion In lung and breast cancer, cases which highly express IL-15 are mostly in early stage of tumor, with small tumor size, without lymph node metastasis, and with more NK cells and CD8+ cells infiltrating in tumor tissue; cases which lowly express IL-15 are mostly in late stage of tumor, with large tumor size, lymph node metastasis, and less NK cells and CD8+ cells infiltrating in tumor tissue. IL-15 may inhibit the development of human lung cancer and breast cancer tumor.
