血竭素对瘢痕成纤维细胞胶原合成的影响

【摘要】 目的:研究天然药物血竭素高氯酸盐(dracorhodinperchlorate,Dp)对瘢痕成纤维细胞合成胶原的影响,为开发新的治疗增生性瘢痕的有效药物提供实验依据。方法:以四甲基偶氮唑(MTT)法、吸光度检测法及逆转录聚合酶链反应(RT-PCR)法分别测定血竭素对成纤维细胞增殖、培养基中的胶原含量以及Ⅰ、Ⅲ型前胶原mRNA表达的影响。结果:MTT法显示血竭素对成纤维细胞的抑制率增高,且呈现浓度和时间依赖性(P<0.05),还能显著降低培养基中可溶性胶原的含量(P<0.05)。RT-PCR法对Ⅰ、Ⅲ型前胶原的mRNA检测显示随着药物浓度增高其表达量降低(P<0.05)。结论:血竭素对瘢痕成纤维细胞的增殖和胶原合成具有显著的抑制作用。

【Abstract】 Objective To study the effect of herbal medicine dracorhodin perchlorate on collagen synthesis in scar-derived fibroblasts and evaluate new drug for human hypertrophic scars. Methods The fibroblasts were cultured as an experimental model and the effect of dracorhodin perchlorate were detected. The cell proliferation was determined by MTT assay, and the collagen production by photometric determination. The mRNA expression of typeⅠand Ⅲ procollagen was detected by RT-PCR. Results It was shown that exposure to dracorhodin perchlorate could increase the inhibiting rate of fibroblasts by MTT assay and dependent on concentration and time(P<0.05), otherwise significantly reduce the soluble collagen content in the culture medium(P<0.05). The results of RT-PCR showed the expression of typeⅠand Ⅲ procollagen was decreased along with the increasing of the concentration of the drug(P<0.05). Conclusion dracorhodin perchlorate could significantly inhibit the proliferation and the collagen synthesis of fibroblasts in vitro.
