HPLC测定天参胶囊中盐酸小檗碱的含量

【摘要】 目的建立了高效液相色谱法测定天参胶囊中盐酸小檗碱含量的方法。方法固定相为Kromasil-C18(250 mm×4.6 mm,5 μm)色谱柱;流动相为V(甲醇):V[1.2%三乙胺溶液(醋酸 40 mL,三乙胺12 mL,加水稀释至1 000 mL)]=45:55;流速为1.0 mL/min,检测波长345 nm。结果天参胶囊中盐酸小檗碱能与多种物质有效分离。盐酸小檗碱在3—15μg/mL范围内线性关系良好,r=0.9990,平均回收率为99.5%(n=5),RSD为2.5%。结论本法简便、快速、准确, 可用于天参胶囊的质量控制。

【Abstract】 Objective To establish a high performance liquid chromatographic method for the determination of berberine hydrochloride in Tianshen capsule. Methods Chromatographic conditions included Kromasikl-C18(250 mmΤ4.6 mm, 5 μm) as stationary phase, the mobile phase was composed of methanol and the mixture of acetic acid-triethylamine-water (40 mL acetic acid and 12 mL triethylamine were mixed and diluted to 1000mL with water)(45:55, v/v). The UV detection wavelength was located at 345 nm with a flow rate of 1.0 mL/min. Results Berberine hydrochloride could be separated completely from other components in Tianshen capsule. Good linear relationship could be obtained in the range of 3~15 μg/mL for berberine hydrochloride(r = 0.9990). The average recovery was 99.5%(n = 5, RSD=2.5%). Conclusion This method is simple, rapid and accurate and can be used for the quality control of Tianshen capsule.
