胎鼠宫内缺血/再灌注损伤后细胞凋亡的分析
Study of fetal kidney cells’ apoptosis after ischemia/reperfusion injury
【摘要】 目的探讨凋亡在急性缺血/再灌注损伤中的作用。方法取孕21 d Wistar大鼠制成宫内缺血/再灌注损伤模型,取胎鼠肾组织行TUNEL分析凋亡细胞指数,免疫组化分析Caspase-3的表达。结果缺血5 min再灌注0 min、2 h、6 h、24 h肾组织凋亡细胞数分别为(12.99±0.18)%、(22.36±0.28)%、(22.73±0.56)%、(32.56±0.62)%和(18.57±0.35)%;再灌注2 h后,随着再灌注时间的延长,凋亡细胞数明显增加,与对照组(12.72±0.16)%比较,有显著差异(P<0.001);凋亡细胞指数以再灌注6h最高;Caspase-3的表达分别为(0.15±0.04),(0.30±0.04),(0.35±0.05),(0.30±0.03),(0.14±0.02),与对照组(0.10±0.03)比较,有显著差异(P<0.05),表达高峰于再灌注2 h出现。结论胎鼠宫内急性缺血再灌注损伤后,出现肾脏细胞不同程度的凋亡,伴有Caspase-3表达增强,Caspase-3表达比TUNEL能更早地反映细胞凋亡。
【Abstract】 Objective To evaluate the role of apoptosis on ischemia/reperfusion injury in rats.Methods Twenty-one day pregnant Wistar rats were used to make model of ischemia/reperfusion injury,fetal kidneys were taken away for TUNEL analysis,the expression of Caspase-3 was analysed by immunohistochemistry technique.Results After ischemia 5min reperfusion 0min,2h,6h,24h,apoptotic cells of kidneys were(12.99±0.18)%,(22.36±0.28)%,(22.73±0.56)%,(32.56±0.62)%,(18.57±0.35)% resepectively,from the time of reperfusion 2h,apoptotic cells increased significantly(P<0.001),compared with the control group(12.72±0.16)%,and the peak was at reperfusion 6h.The expression of Caspase-3 were(0.15±0.04),(0.30±0.04),(0.35±0.05),(0.30±0.03),(0.14±0.02) respectively,differences were significant compared with the control(0.10±0.03),(P<0.05).The peak appeared at reperfusion 2h.Conclusion After acute ischemia/reperfusion injury,fetal rat’s kidney apoptotic cells were increased,so as to the expression of Caspase3.The expression of Caspase-3 can reflect apoptosis earlier than TUNEL.
