兔眼玻璃体腔注射万古霉素对视网膜谷氨酸含量的影响

【摘要】 目的观察兔眼玻璃体腔注射万古霉素后视网膜谷氨酸含量变化,初步探讨毒性反应机制。方法48只兔随机分为3个实验组和1个对照组,每组12只。分别向3个实验组兔眼玻璃体腔注入1、25、mg/0.1 mL的万古霉素溶液,对照组注射生理盐水0.1 mL。术后1、4、71、4 d采用比色法检测视网膜内谷氨酸的浓度,进行统计分析。结果与对照组比较,2 mg组视网膜谷氨酸含量于4~7 d升高明显(P<0.05),5 mg组各时间点均升高明显(P<0.05),7 d达高峰,之后逐渐下降。剂量因素对视网膜谷氨酸含量的作用显著(P<0.01),时间因素对谷氨酸含量的作用不显著。结论兔眼玻璃体腔注射万古霉素超过安全剂量,可引起视网膜内谷氨酸含量升高,本实验将有助于抗生素所致视网膜毒性反应机制的研究。

【Abstract】 Objective To explore the changes of glutamate levels in retina of rabbits’ eyes after intravitreal injection with vancomycin,and discuss the mechanism of retinal toxicity.Methods Forty eight rabbits were randomized to 4 groups with 12 rabbits in each group.0.1 mL vancomycin in doses of 1mg,2mg and 5mg was intravitreally injected into the eyes of treated groups rabbits respectively.0.1 mL saline solution was injected into the vitreous body of the rabbits in the control group.Concentration of glutamate in retina were determined with UV-VIS Spectrophotometer on 1st,4th,7th and 14th day.Results Compared with the control group,the glutamate levels in the retina was obviously increased in 2 mg group(4~7 d,P<0.05),and in 5 mg group(all point-in-time,the pinnacle was at 7th day,P<0.01).The effect of time factor on the changes of glutamate levels in retina is not significant,but of dose factor is significant(P<0.01).Conclusion The doses of vancomycin that is intravitreally injected more than 2 mg can increase the glutamate levels obviously in retina of rabbits’ eyes.The article will be helpful to explore the mechanism of retinal toxicity which is resulted from antibiotics.
