酶法测定大麦提取物β-葡聚糖含量研究

【摘要】 目前国际认可β-葡聚糖含量测定方法是酶法,此法测定成本高,我国目前多采用苯酚- 硫酸法测定,但测定结果准确性较差。该研究在国际β-葡聚糖酶法测定基础上用纤维素酶代替昆布多糖水解酶水解β-葡聚糖,用苯酚-硫酸法代替葡萄糖试剂盒法测定β-葡聚糖酶解后得到葡萄糖含量,设计适于我国应用的β-葡聚糖酶法测定方法。此法测定过程为:1.5 ml浓度为60μg/mL 标准β-葡聚糖和一定浓度样品,用浓度为50U/mL纤维素酶0.5 ml酶解60 min;然后用蒸馏水稀释至30 ml,从中吸取0.5 ml到另一试管,再加入浓度为2 U/mL β-葡聚糖酶1ml,作用15 min后,用苯酚-硫酸法测定标准β-葡聚糖和样品酶解液中葡萄糖含量,计算出样品中β-葡聚糖含量。

【Abstract】 Enzyme method to determinate β-glucan was advised internationally with its high cost, phenol-sulphuric acid method was mainly used in our country with its poor accuracy. The determination method applicable to our country was established at base of enzyme method with change of Lichenase by cellulose enzyme and change of glucose assay kit by phenol-sulphuric acid method. The new enzyme method procedure was: 1.5 mL β-glucan (60 μg/mL )and sample were hydrolyzed with 0.5 mL cellulose enzyme(50 U/mL)for 60 min; then were diluted 30 mL, 0.1 mL β-Glucosidase(2 U/mL)were added in 0.5 mL hydrolysate, glucose content of hydrolysate from standard β- glucan and sample was determined by phenol-sulphuric acid method after 15 min, content of β-glucan in sample was calculated out.
