颈动脉硬化斑块环氧化物酶-2表达及其干预的实验研究
Effects of TONGXINLUO capsule on the expression of COX-2 and mPGES-1 in the rabbit carotid artery atherosclerotic plaque
【摘要】 目的探讨通心络对兔颈动脉粥样硬化斑块环氧化物酶-2(COX-2)及Ⅰ型膜相关前列腺素E合成酶(mPGES-1)表达的影响。方法采用32只雄性新西兰大白兔,其中8只为正常对照组(1组),另24只建立兔颈动脉粥样硬化性狭窄的动物模型。将模型兔随机分为颈动脉狭窄无干预组(2组)、塞来昔布治疗组(3组)、通心络治疗组(4组),每组各8只。药物干预2周后取狭窄段颈动脉及对侧相应段的颈动脉,行逆转录聚合酶链反应及Western印记(Westernblot)检测颈动脉斑块药物干预后COX-2及mPGES-1表达。结果2、3、4组颈动脉粥样硬化模型兔斑块的COX-2mRNA、mPGES-1mRNA及蛋白表达上调水平,与1组比较差异有显著性意义(P<0.05),3组、4组较2组下调水平差异有显著性意义(P<0.05),3组较4组下调更显著(P<0.05)。结论通心络可能有益于抗颈动脉粥样硬化斑块形成的作用。
【Abstract】 Objective To explore the effect of TONGXINLUO capsule on the expression of cyclooxygenase type 2(COX-2) and membrane associated prostagladin E synthase-1(mPGES-1) in the rabbit carotid artery atherosclerotic plaque.Methods After establishing carotid artery scleratheroma models in rabbits.The models were randomly divided into three groups: carotid artery atherosclerotic plaque group(group 2);TONGXINLUO therapic group(group 3);celecoxib therapic group(group 4);control group (group 1). COX-2 and mPGES-1 expression of the carotid arteries was evaluated by RT-PCR and western blot.Results There was more significant downregulation of COX-2 and mPGES-1 gene expression (P<0.05) in group 3 and group 4 than in group 2 after TONGXINLUO capsule and celecoxib therapy in the rabbit models.Conclusion TONGXINLUO capsule may have the action of protecting carotid artery against atherosclerotic plaque formation in the rabbit models.
