复方芪参降脂饮抗实验性动脉粥样硬化作用及其可能机制

【摘要】 目的探讨复方芪参降脂饮对实验大鼠动脉粥样硬化(AS)的治疗作用及其可能机制。方法SD大鼠24只,随机分为对照组(普通饲料)、模型组(高脂饲料)、中药组(高脂饲料,同时给以复方芪参降脂饮灌胃);每组8只。8 wk后采血,测血清TC、TC、LDL-C和HDL-C水平,测定血清丙二醛(MDA)水平及超氧化物歧化酶(SOD)活性。处死SD大鼠取其主动脉,观察主动脉组织形态学变化,用免疫组化和Western blot检测主动脉壁细胞间黏附分子-1(ICAM-1)蛋白表达水平。结果中药组主动脉内皮损伤明显轻于模型组,中膜平滑肌排列也较模型组整齐。中药组血脂和MDA水平明显低于模型组(P<0.01),而SOD活性则明显高于模型组(P<0.05)。模型组大鼠主动脉壁ICAM-1蛋白表达量明显高于中药组(P<0.01)。结论复方芪参降脂饮能够抑制实验大鼠动脉粥样硬化的发生、发展,其作用机制可能是降低血脂、清除氧自由基以及抑制ICAM-1在血管壁的表达。

【Abstract】 AIM To study the effects and related mechanism of complex prescription Qishenjiangzhiyin on the rat model of atherosclerosis. METHODS Twenty-four male SD rats were randomly and equally divided into control group (fed with normal diet) ,model group (fed with high cholesterol diet) and treatment group (fed wieh high cholesterol diet plus complex prescription Qishenjiangzhiyin);There were eight rats in every group. Eight weeks later, blood was collected . Blood-fat level, serum malondialdehyde (MDA) concentrations and superoxide dismutase (SOD) activity were measured . SD rats were killed and the aortas were collected for histomorphology studies. The intercellular adhesion molecule-1(ICAM-1) expression of aorta was measured by immunohistochemistry and Western blot. RESULTS The endothelial damage of the aortas in treatment group was less severe than that in model group. The smooth muscle cell of the aortas in treatment group lined up in orderer than that in model group,too.The level of blood-fat and serum MDA in the treatment group decreased significantly as compared with that of model group(P<0.01) .Serum SOD activity increased in treatment group as compared with that of model group(P<0.05). The ICAM-1 expression in aortas in treatment group was significantly lower than that in model group(P<0.01). CONCLUSION Complex prescription Qishenjiangzhiyin can inhibit the occurrence and development of atherosclerosis in rats. This effects may be achieved by decreasing the blood-fat, eliminating oxygen-derived free radicals and down regulating the expressions of ICAM-1 in aorta.
