【摘要】 目的:评价国产吉西他滨联合氟尿嘧啶(5FU)/甲酰四氢叶酸(CF)组成的GLF双周方案作为二线方案治疗晚期乳腺癌的疗效和毒性。方法:34例既往曾接受蒽环类和/或紫杉类方案化疗的晚期乳腺癌患者,采用GLF双周方案,即吉西他滨1200mg/m2,静脉输注,CF200mg/m2,静脉输注,5FU325mg/m2先静脉推注,然后2.8g/m2持续48小时滴注;每2周重复1次,每2次为1个周期。结果:34例中CR2例,PR13例,NC11例,PD8例,总有效率为44.1%。Ⅲ～Ⅳ度白细胞下降为17.6%,Ⅲ度血小板下降为5.9%,Ⅲ度粘膜炎5.9%,其他不良反应轻微。结论:国产吉西他滨联合5FU及CF组成的GLF双周方案作为二线方案治疗对蒽环类和/或紫杉类方案耐药的晚期乳腺癌疗效令人满意,不良反应少,值得进一步研究。更多还原

【Abstract】 Objective;To evaluate the efficacy and toxicity of semimonthly gemcitabine plus 5-fluorouracil/leucovorin in the treatment of advanced breast cancer resistant to anthracyclines and/ or taxanes. Methods:Thirty four cases of advanced breast cancer pretreated with anthracyclines and/or taxanes were enrolled into this study. Gemcitabine 1 200mg/m2 i.v, combined with leucovorin 200mg/m2 i.v, then 10 minute intravenous bolus of 5-FU 325mg/m2 followed by 48 hour infusion of 5-FU 2.8g/m2 using an ambulatory pump. The regimen was given per 14 days. One cycle consisted of twice chemotherapy regimens. All enrolled patients received at least two cycles of treatment. Results:It showed that CR 5.9%, PR 38.2%, NC 32.4%, PD 23.5% with an overall response rate of 44.1%. The main side effect included grade Ⅲ～Ⅳ leukopenia 17.6%, grade Ⅲ thombocytopenia 5.9%. Conclusion:Semimothly gemcitabine plus 5-fluorouracil/leucovorin (GLF regimen) is effective for advanced breast cancer resistant to anthracyclines and/or taxanes, and toxicities are well tolerated.更多还原
