三七总皂甙对大鼠脑出血模型中神经细胞凋亡、Bcl-2表达的影响

【摘要】 目的:研究大鼠实验性脑出血模型中神经细胞凋亡发生的规律以及与Bcl-2表达的关系,观察三七总皂甙对出血性脑损伤的保护作用。方法:将雄性SD大鼠随机分成正常组、假手术组、脑出血模型组、三七总皂甙治疗组。采用Ⅶ型胶原酶注入到大鼠右侧苍白球制备脑出血模型,每组分别在术后6 h、1 d、3 d、5 d 4个时间点取SD大鼠脑组织标本,应用末端脱氧核苷酸转移酶介导的脱氧三磷酸尿苷缺口末端标记法(TUNEL)检测细胞凋亡,免疫组化法检测Bcl-2蛋白表达。结果:正常组与假手术组比较,大鼠脑出血后各时间点脑组织TUNEL阳性细胞数、Bcl-2表达显著增高(P<0.01);与脑出血模型组比较,三七总皂甙治疗组1 d、3 d、5 d TUNEL阳性细胞数显著减少(P<0.05-0.01),Bcl-2显著增高(P <0.05-0.01)。结论:细胞凋亡参与了脑出血后神经细胞的继发损伤过程,Bcl-2有抑制细胞凋亡的作用;三七总皂甙可以增高脑出血后Bcl-2的表达,对脑组织具有保护作用。

【Abstract】 Objective:It is to study the characteristics of nerve cell apoptosis and influence of SanQiZongZaoDai on the expression of Bcl-2 in rats with intracerebral hemorrhage. Methods:All Sprague - Dawley male rats were randomly divided into normal group (group A), pseudo -operation group (group B), model group (group C) and SanQiZongZaoDai treatment group (group D). ICH models were induced in rats by injection ofⅦtype collagenase into the right globas pallidus. The brain tissue specimen in each group were gathered at 6h、1d、3d and 5d after operation,respectively Then the apoptosis was detected by terminal deoxynucleotidy transferase mediated d UTP nck end labeling (TUNED and the expression of Bcl-2 protein was detected by immunochemical attaining in rat brain after ICH. Results:Compared with group A and group B,the expression of TUNEL and Bcl-2 in brain tissue in group C were obviously increased (P<0.01) . Compared with group C, the expression of TUNEL in brain tissue in group D was obviously reduced (P<0.05-0.01) and the expression of Bcl-2 was obviously decreased (P<0.01) in 6h、1d、3d and 5d . Conclusion:SanQiZongZaoDai can decrease the expression of Bcl-2 in rat after ICH, which may be related with its anti- apoptosis effect following ICH.
