γ-氨基丁酸转运体-1与局灶性脑缺血再灌注神经元损伤关系的实验研究

【摘要】 目的探讨γ氨基丁酸转运体1(GAT1)与脑缺血再灌注神经元损伤的关系。方法制备大鼠局灶性脑缺血再灌注模型,分别在脑缺血再灌注3h、6h、12h、24h及3d取损伤侧脑组织进行免疫组化染色,测定标本中GAT1阳性神经元数目和光密度值,并与正常组和假手术组比较。结果脑缺血再灌注3hGAT1表达明显上调(P<0.05),6h达到高峰(P<0.01),12h开始下降,24h后与对照组比较差异无显著性。结论脑缺血再灌注早期GAT1阳性神经元明显增多,其在局灶性脑缺血再灌注中可能参与神经元损伤病理生理过程。

【Abstract】 Objective To study the relationship between GAT-1 and neuron injury in focal cerebral ischemia/infusion model.Methods Focal cerebral ischemia/reperfusion models were established in rats by thread embolic, then the brain tissues were taken at 3 h, 6 h,12 h, 24 h and 3 d after cerebral ischemia/reperfusion.The number of GAT-1 positive neurons were calculated and the average optical density were detected by immunohistochemical technique in different time points after cerebral ischemia/reperfusion, compared with the normal group and the sham operated group.Results The expression of GAT-1 was up-regulation obviously at 3 h after cerebral ischemia/reperfusion( P<0.05), reached a peak at 6 h( P<0.01), begun to decrease at 12 h,there was no significant different from control group at 24 h.Conclusions The number of GAT-1 positive neurons increase in the early stage after cerebral ischemia/reperfusion,which may involve in the pathological progress of neuron injury.
