外周炎症时大鼠背根神经节细胞μ-和κ-阿片受体mRNA的表达
Expression of μ-and κ-opioid receptor mRNA in rat dorsal root ganglion neurons during peripheral inflammation
【摘要】 目的观察外周炎症时大鼠背根神经节(dorsalrootganglion,DRG)细胞μ-和κ-阿片受体mRNA表达的变化。方法用角叉菜胶(carrageenan,CAR)在大鼠右后足皮下注射产生外周炎症,分别于正常和炎症后6、12、24和72h取右L4~5DRG。用原位杂交的方法观察正常及炎症期间DRG细胞μ-和κ-阿片受体mRNA表达的变化。结果正常大鼠的部分DRG细胞内可见μ-和κ-阿片受体mRNA表达。外周炎症后6h,表达μ-和κ-阿片受体mRNA的DRG细胞的数和μ-和κ-阿片受体mRNA表达量增加,24h表达量最高,72h下降至正常。结论外周炎症能使大鼠DRG细胞μ-和κ-阿片受体mRNA表达增加。提示外周炎症时大鼠DRG细胞μ-和κ-阿片受体合成增加。为阿片类物质应用于外周镇痛提供理论依据。
【Abstract】 Objective To observe the changes of μ-and κ-opioid receptor mRNA expression in rat dorsal root ganglion neurons(DRG)during peripheral inflammation.Methods Peripheral inflammation was induced by injection of carrageenan into the plantar surface of the right hindpaw of rats. The right L_ 4-5 DRG was isolated taken at 6, 12, 24,and 72h after inflammation . The expression levels of μ-and κ-opioid receptor mRNA in normal and inflamed rat DRG neurons were detected by in situ hybridization.Results There were expressions of μ-and κ-opioid receptor mRNA in part of normal rat DRG neurons . The expression levels of μ-and κ-opioid receptor mRNA were increased at 6h after inflammation, and reached the peak level at 24h after inflammation, then decreased to the normal at 72h after inflammation. Conclusion During peripheral inflammation,the expressions of μ-and κ-opioid receptor mRNA were increased in rat DRG neurons,suggesting that μ-and κ-opioid receptor of rat DRG neurons synthesis is increased during peripheral inflammation.
