【摘要】 目的:研究凋亡抑制蛋白Livin在膀胱移行细胞癌(BTCC)组织中的表达情况,探讨Livin蛋白与BTCC发病、演进和复发的关系。方法:应用半定量逆转录-聚合酶链反应(RT-PCR)方法检测30例BTCC组织和10例正常膀胱组织新鲜标本中Livin基因mRNA的表达情况。结果:10例正常膀胱组织Livin基因均无表达,20例膀胱癌组织标本Livin基因为表达阳性(20/30,66.6%),且同时表达Livinα和Livinβ两种亚型。Livin基因是BTCC发生和演进中的早期事件之一,在BTCC早期(Ta+T1)和低分级阶段(G1),Livin有较高的阳性表达率,分别达71.4%和69.2%。Livin基因mRNA表达与BTCC分级和分期无关,但与其是否复发密切相关,复发组Livin阳性率为85.0%,远高于未复发组的16.7%。结论:Livin基因与BTCC的演进和复发有关,可作为BTCC早期诊断和评判术后高复发风险的一项重要指标。更多还原

【Abstract】 Objective:To study the expression of Livin,an apoptosis inhibitor,in tissues of transitional cell carcinoma of baldder and to explore its clinical significance.Methods:Semiquantitative reverse transcription polymerase chain reaction（RT-PCR） was used to investigate the mRNA expression of Livin gene in 30 bladder transitional cell carcinoma samples and 10 normal samples from urinary bladder.Results:None of all the 10 normal bladder samples was detected Livin,whereas,expression of livin in 20 carcinoma samples was positive（20/30,（66.6）%）（both α and β isoform）.Livin was found positive in（71.4）% of the T_a/T₁ group,and（69.2）% in the grade 1 group,respectively.The expression of Livin gene was not correlated with the stage and grade of BTCC,whereas,the presence of Livin was associated with tumors with high risk of relapse.The positive expression of Livin mRNA in recurrent cases was higher than that of non-recurrent cases,the former was（85.0）%（17/20） and the latter was（16.7）%.Conclusions:Positive expression of Livin in bladder transitional cell carcinoma is associated with high risk of relapse.更多还原
