持续性脊髓压迫伤血管内皮生长因子mRNA表达的变化
Expression of vascular endothelial growth factor-mRNA in the rats′spinal cord continuous compression injury
【摘要】 目的研究持续性脊髓压迫伤血管内皮生长因子(VEGF)mRNA表达的变化,并探讨其在脊髓压迫伤中表达的作用和意义。方法健康封闭群Wistar大鼠60只,采用平头塑料螺钉后路压迫复制大鼠持续性脊髓压迫伤的动物模型(脊髓50%压迫)。以改良Tarlov评分、BBB-21点评分及斜板实验评价后肢运动功能;应用原位杂交技术检测脊髓组织在持续性压迫适应中VEGF-mRNA的表达,并结合计算机图像分析仪进行定量分析。结果持续压迫各时间组大鼠后肢功能明显减弱,在2d组大鼠后肢功能进一步恶化,随着持续压迫时间的推移,大鼠脊髓运动功能有一小幅度的自发性恢复,在持续压迫3周左右恢复较明显。假手术组未见VEGF-mRNA免疫阳性细胞。脊髓压迫伤后1d,VEGF-mRNA表达明显上调(P<0·01),2d左右达高峰,1周时表达明显下调(P<0·05),3周时见到零星VEGF-mRNA免疫阳性细胞。结论在持续性脊髓压迫损伤的发生与发展中,VEGF-mRNA的表达可能参与了脊髓组织的保护及损伤后的修复。
【Abstract】 Objective To investigate the expression changes and effects of vascular endothelial growth factor (VEGF) gene in rats′spinal cord in different phases of spinal cord continuous compression injury development. Methods The spinal cord continuous compression injury model was made by compression to the posterior spinal cord using a blunt plastic screw. Sixty Wistar male rats were randomly divided into compression group and control operative group. Changes in the behavior and limb function experimental rats were observed. The expression changes of VEGF-mRNA was detected through in situ hybridization methods. The VEGF-mRNA positive cells were quantitatively analyzed by a computer image analysis system. Results The limb function of experimental rats significantly weakened,further deteriorated at 2 d group,and slightly recovered after 1 week.The expression of VEGF-mRNA was not detected in control operation group,but significantly increased at 1 d(P<0.01) and peaked at 2 d in the compression group. Then,it decreased significantly at 1 week (P<0.05),and disappeared at 3 weeks. Conclusions VEGF gene expression might play a role in neuronal protection and repair following spinal cord continuous compression injury.
