【摘要】 探讨HBV感染者外周血单个核细胞(PBMC)中HBVDNA的检出情况,观察拉米夫定抗病毒治疗后PBMC中HBVDNA的变化。常规分离65名HBV感染者和10名健康体检者的PBMC,裂解PBMC抽提总DNA,并进行HBVDNA的定量检测。其中7名HBV感染者进行拉米夫定抗病毒治疗。HBVDNA阳性者其PBMC中HB- VDNA的检出率为90．2%(46/51),HBVDNA阴性者中检出率为7．1%(1/14)。7例HBVDNA阳性的慢性HBV感染者经拉米夫定抗病毒治疗后,血清及PBMC中HBVDNA定量均逐渐降至检测水平以下,PBMC中HBVDNA的消失较血清无明显延迟(P＞0．05)。HBV感染者其PBMC有较高的HBV感染率。拉米夫定对血及PBMC中HBVD- NA均有明显的抑制作用。更多还原

【Abstract】 To detect HBVDNA in PBMCs from patients infected with HBV,and to observe the changement of HBVDNA in both serum and PBMCs after treated with lamivudine.PBMCs of 65 cases infected with HBV and l0 healthy donors were isolated,and then lyzed.Total DNA were extracted.HBVDNA was detected by quantitative PCR assay.Among those pa- tients,7 cases were treated with lamivudine.The detection rate of HBVDNA in PBMCs from patients with HBVDNA posi- tive was 90.2%(46/51),while that in PBMCs from patients with HBVDNA negative was only 7.1%(1/14).Compared with serum HBVDNA,the disappear of HBVDNA in PBMCs was not postponed.The HBVDNA positive rate was high in patients infected with HBV.Lamivudine can inhabit HBV replication effectively in both serum and PBMCs.更多还原
