高效液相色谱法测定乳酸羟乙酸共聚物微球中雌二醇的含量

【摘要】 建立了高效液相色谱法测定雌二醇缓释微球载药量的方法。色谱柱为C18柱,流动相为甲醇-0.01mo·lL-1磷酸缓冲液(pH7.0)-三乙胺(70:30:0.2,v/v/v),检测波长为280nm。结果表明在(5.04—80.64)μg·mL-1浓度范围内,峰面积对浓度有良好的线性关系。日内RSD≤0.98%,日间RSD≤1.12%,回收率为99.34%,这说明了HPLC法专属性强,结果准确。

【Abstract】 An HPLC method was established to determination the content of estradiol-loaded Poly(lactic-co-glycolic acid) microspheres. The HPLC method with C18 column, the mobile phase of KH2PO4 (pH 7.0; 0.01 mol) / methanol/ triethylamine (70:30:0.2 v/v/v) and the detection wavelength of 280 nm was used. The method had a good linear relationship in the range of (5.04~80.64) μg·mL-1, the precision was within-day RSD≤0.98% and day-to-day RSD≤1.12% respectively, the average recovery was 99.34%. All of these showed that the proposed method was selective and accurate.
