防龋DNA疫苗pGJA-P/VAX免疫小鼠后的抗DNA抗体检测

【摘要】 目的:检测防龋DNA疫苗pGJA-P/VAX经鼻腔免疫和肌肉免疫后是否会产生抗DNA抗体。研究该疫苗是否会导致自身免疫性疾病。方法:提取纯化的质粒pGJA-P/VAX分别经鼻腔和肌肉免疫小鼠,于第0、2、4、8周取血清做抗DNA抗体检测实验(ELISA法)。结果:各实验组之间均无显著性差异,各组免疫前后及各组间比较(P>0.01)。结论:说明DNA疫苗pGJA-P/VAX及其载体pVAX1对小鼠均不会引起自身免疫反应,导致自身免疫病。

【Abstract】 Objective: To compare the levels of anti-ds-DNA antibodies and anti-ss-DNA antibodies in mice immunized with pGJA-P/VAX and mice without such treatment.Methods: pGJA-P/VAX was administrated imtramuscularly or intranasally to mice,and all mice received a booster immunization 2 weeks later after the initial immunization.The concentrations of anti-ssDNA antibodies and anti-dsDNA antibodies of unimmunized mice were measured by the method of ELISA,which were used as baseline for the comparison study.2,4 and 8 weeks after the initial immunization,blood samples of these immunized mice were collected to check the levels of anti-ssDNA antibodies and anti-dsDNA antibodies by using the same method.Results: No statistically significant changes of anti-ssDNA antibodies and anti-ds-DNA antibodies were found between unimmunized and immunized mice.Conclusion: The blood levels of anti-ss-DNA antibodies and anti-ds-DNA antibodies did not statistically significantly increase by the repeated and high dose adminstration of anticaries DNA vaccine pGJA-P/VAX.Additionally,autoimmune diseases were found among the immunized mice at the specific experiment condition.
