【摘要】 目的研究COX-2在人大肠腺癌发生发展过程中的作用,并探讨其与大肠腺癌转移的关系.方法采用免疫组化SP法,观察人大肠腺瘤30例、无淋巴结转移的大肠腺癌35例、有淋巴结转移的大肠腺癌35例组织中COX-2蛋白的表达和定位特点.结果COX-2蛋白的表达定位于细胞浆和细胞膜,从大肠腺瘤到大肠腺癌其蛋白表达明显增强,差异具有统计学意义(P=0.000);在无淋巴结转移的大肠腺癌和有淋巴结转移的大肠腺癌原发灶之间,其蛋白表达差异有统计学意义,且呈正相关关系(P=0.035,Spearm an相关系数=0.253);从高分化大肠腺癌到低分化大肠腺癌之间,其蛋白表达明显增强(P=0.009).结论COX-2蛋白表达的上调在大肠腺癌的发生发展过程中可能起着重要作用,且COX-2可作为大肠腺癌患者预后判断的良好指标之一.更多还原

【Abstract】 Objective To investigate the effect of COX-2 protein in the carcinogenesis,progression,and metastasis of human colorectal adenocarcinomas.Methods COX-2 protein was detected by SPimmunohistochemistry in formalin-fixed and paraffin-embeded tissues of 30 colorectal adenoma tissues,35 colorectal adenocarcinoma tissues with no regional lymph nodes metastasis,and 35 primary colorectal adenocarcinoma tissues with regional lymph nodes metastasis.Results The expressions of COX-2 protein distributed in cytoplasms and cell membranes,it was gradually increased from colorectal adenoma tissues to colorectal adenocarcinoma tissues(P=0.000).From the well differentiated colorectal adenocarcinoma tissues to the poor differentiated tissues the expressions of COX-2 protein were significantly increased(P=0.009).And the expressions of COX-2 protein between colorectal adenocarcinoma tissues with no regional lymph nodes metastasis and primary colorectal adenocarcinoma tissues with regional lymph nodes metastasis had significantly difference(P=0.035).The expressions of COX-2 were not correlated with some clinicopathological indices(P>0.05),such as gender,age,site of colorectal adenocarcinoma,et al.Conclusions COX-2 can be regard as markers for predicating the metastasis and invasion of the colorectal adenocarcinoma.So it was a well prognostic parameter for colorectal cancer.更多还原
