天麻醒脑胶囊对学习记忆功能的影响及机理研究

【摘要】 目的评价天麻醒脑胶囊对动物学习记忆功能的影响及其初步作用机理.方法采用东莨菪碱、乙醇及亚硝酸钠复制小鼠和大鼠记忆障碍模型,运用避暗法研究天麻醒脑胶囊对动物学习记忆功能的影响,同时探讨其对脑组织乙酰胆碱酯酶(AChE)活性、丙二醛(MDA)及脂褐质水平的影响.结果高、中剂量(大鼠:高剂量1.2 g/kg,中剂量0.6 g/kg;小鼠:高剂量2.4 g/kg,中剂量1.2 g/kg)的天麻醒脑胶囊能明显减少给药组大鼠及小鼠5 m in内的错误次数,延长触电潜伏期.高、中、低3个剂量的天麻醒脑胶囊均可减少大鼠脑组织MDA水平,降低AChE活性,同时明显抑制脑组织脂褐质含量的升高.结论天麻醒脑胶囊通过多种途径改善记忆损伤模型动物的学习记忆功能,其机制可能与降低脑组织AChE活性,降低脑组织中脂质过氧化物、脂褐质等的含量有关.提示天麻醒脑胶囊具有良好的促智作用.

【Abstract】 Objective To study the effects of Timaxingnao Capsule on the abilities of learning and memory and its preliminary mechanisms.Methods ICR mice and SD rats were respectively induced by scopolamine hydrobromide,alcohol,and NaNO2 to make models.The test of avoidance response to darkness was used.The influence of Timaxingnao Capsule was investigated on acetylcholine esterase(AChE),malondiadehyde(MDA) and lipofuscin in brain tissue.Results In the test of avoidance response to darkness,Tianmaxingnao Capsule(1.2 and 2.4g/kg for mice,0.6and 1.2g/kg for rats) could significantly reduce the error times within 5min in dysmnesia groups(P< 0.01,P< 0.05) and prolong the incubation periods of memory.The three doses of Tianmaxingnao Capsule decreased the level of MDA,suppressed the activity of AChE and obviously inhibited the increase of the lipofuscin in rat brain tissue.Conclusion Tianmaxingnao Capsule could improve the abilities of learning and memory in animal models through multiple ways.The preliminary mechanisms may be associated with its effects on decreasing the activity of AChE,promoting the function of cerebral monoamine nervous system in brain tissue and reducing the level of lipid peroxidation in brain tissue.It is suggested that Tianmaxingnao Capsule have the effects of improving learning and memory.
