大黄鱼线粒体DNA的限制性内切酶图谱
Studies on Restriction Enzyme Map of Mitochondrial DNA from Pseudosciaena crocea(Richardson)
【摘要】 从大黄鱼(Pseudosciaena crocen)肝脏中提取线粒体DNA(mtDNA)。用限制性核酸内切酶酶切后,得到酶切图谱。以1 kb ladder作为相对分子量标准,以电泳迁移率为纵坐标,分子量的对数值为横坐标作标准图,利用电泳迁移率与分子量的对数值的线性关系,计算出各酶切片段的分子量,并推算出大黄鱼mtDNA分子量为27.33×10~6 D,大小为16.891 kb左右。根据单酶切和双酶切的数据,以及mtDNA是环状分子等特征,建立了大黄鱼的mtDNA酶切图谱。
【Abstract】 The mtDNA from liver of Peudosciaena crocea has been isolated and purified by the method of density gradi- ent centrifugation and DnaseⅠ,Rnase degestion,and has been analyzed with 7 kinds of restriction endonuclease.The number of recognition site of EcoRⅠ、HindⅢ,PstⅠ,BglⅡ,BamHⅠ,BgⅠand XbaⅠon this mtDNA is 1(2)、4、2、2 (3)、2、1、and 3 respectively.The molecular weight of Peudosciaena crocea mtDNA is 27.33x 10~6 doltans,about 16.891 kilobase pairs.Simuhanously,we established the restriction map of this mtDNA according to the number and the size of those fragments obtained by single and double digestions.
