骨髓基质细胞的体外培养及分化为成肌样细胞的实验研究
In vitro culture and differentiation of bMSCs transfected by Ad-MyoD gene into myoblast-like cell cells
【摘要】 目的:评估骨髓基质细胞(bMSCs)体外分化为成肌样细胞的能力,探讨骨髓基质细胞作为肌肉组织工程新的种子细胞的可能性。方法:体外分离培养兔骨髓基质细胞,经腺病毒介导的MyoD(Ad-MyoD)基因转染后,观察细胞的形态变化及增殖情况,并行成肌细胞标志物的检测。结果:转染后细胞形态发生明显变化,可见肌管样结构;myogenin及desmin免疫细胞化学染色呈阳性。结论:体外培养的骨髓基质细胞可以向成肌方向转化,有望成为肌肉组织工程新的种子细胞。
【Abstract】 Objective: To assess the ability of inducing bone marrow stromal cells(bMSCs)into Myoblast-like cells, and to approach the possibility of bMSCs as a new seed cells for muscle tissue engineering. Methods: Bone marrow stromal cells of rabbits were separated and cultured in vitro. After being transfected by Ad-MyoD gene, morphlogic change and proliferation of bMSCs were observed and Myoblastic specific marker was examined. Results: After the cells were transfected , there was a significant changes in the cells’ shape. We can see the mytobe-like structure.And the cells were faint positive with immunocytochemical stain of myogenin and desmin. Conclusion: bMSCs have the capability to be differentiated into myoblast and to be regarded as the seed cells for muscle tissue engineering.
