银杏平颤方对帕金森病鼠多巴胺神经元丢失和细胞凋亡的影响

【摘要】 目的探讨银杏平颤方防治帕金森病的机制。方法采用腹腔注射MPTP建立小鼠帕金森病模型。分别应用免疫组化方法和末端脱氧核苷酸转移酶介导的dUTP缺口末端标记法观察中药对中脑黑质多巴胺神经元丢失和细胞凋亡的影响。结果模型组小鼠中脑黑质致密部酪氨酸羟化酶阳性神经元的数目比正常组明显减少,28天比14天减少更明显;银杏平颤方及其拆方各中药治疗组可部分减少黑质致密部TH阳性神经元的丢失;模型组小鼠中脑黑质致密部凋亡细胞的数量明显增多,14天比28天增加更明显,中药各治疗组可不同程度地抑制该部位细胞的凋亡。结论细胞凋亡可能是帕金森病小鼠中脑多巴胺神经元丢失的主要方式之一,银杏平颤方可通过抑制细胞凋亡减少多巴胺神经元的丢失,达到防治帕金森病的作用。

【Abstract】 To investigate the mechanism of Yinxingpingchuan Formula in prevention and treatment of Parkinson’s disease(PD).Methods To establish the mouse model of PD by intraperitoneal injection of MPTP.Immunohistochemistry and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling(TUNEL) technique were used to observe the influence of Chinese herbal drugs on the loss of(mesencephalic) nigra dopamine(DA) neurons and apoptosis.Results In the model group the positive neurons of tyrosine hydroxylase(TH) in mouse nigra compact part decreased obviously than that of the normal group,which was more obvious on 14th day than on 28th day.Yinxingpingchuan Formula and its decompositions could reduce partially the loss of TH positive neurons in mesencephalic nigra compact part.In the model group the apoptosis cells in mouse mesencephalon increased obviously,which was more obvious on 14th day than on 28th day.In the different groups treated with Chinese herbal drugs the apoptosis was inhibited at different degrees.Conclusion Apoptosis maybe one of the common styles of DA neuron loss in mesencephalon of PD mice.Yinxingpingchuan Formula can prevent and treat PD by inhibiting apoptosis and reducing DA neuron loss.
