共轭亚油酸对肥胖大鼠肝脏脂质代谢的影响
EFFECT OF CONJUGATED LINOLEIC ACID ON LIVER LIPID METABOLISM IN OBESE RATS
【摘要】 目的研究共轭亚油酸(CLA)对高脂高糖饮食诱导的肥胖大鼠体质量、血脂及肝脏脂质代谢的影响。方法成年雌性Sprague-Dawley大鼠35只按体质量随机分为5组,即基础对照组、肥胖对照组和CLA低、中、高剂量组(分别为0.4、0.81、.6 g.kg-1)。除基础对照组喂饲基础饲料外,其余4组喂饲高脂高糖饲料,喂饲5周后灌胃CLA 1个月,测定体质量和血脂水平,并对肝脏组织切片进行油红O染色。结果与肥胖对照组相比,CLA低、中、高剂量组体质量分别降低了5.28%、5.67%、7.16%,CLA 3个剂量组的脂体比分别降低了52.8%、54.6%、54.6%(P<0.05);CLA 3个剂量组的血清甘油三酯水平分别降低了18.6%、34.8%、49.1%,其中高剂量组与肥胖对照组相比差异有显著性(P<0.05);CLA组大鼠肝脏切片油红O染色后肝细胞内的红染脂滴随CLA剂量的增加逐渐减少。结论CLA可降低大鼠体质量、血脂水平,并通过影响肝脏脂质代谢起到抗肥胖的作用。
【Abstract】 Objective To study the effect of conjugated linoleic acid(CLA) on body weight,serum lipids level,and liver lipid metabolism in obese rats fed on high-fat and high-carbohydrate diet.Methods Thirty-five female SD rats were divided into five groups:basic control group,obesity control group,and low,medium and high dose of CLA groups(0.4,0.8,1.6 g·kg-1).Except basic control group,the other four groups were fed on high fat and high carbohydrate diet.After feeding for five weeks,the three CLA groups were given different doses of conjugated linoleic acids for one month.Both body weight and serum lipid level were measured,and liver slides were stained with Oil Red O.Results Body weight decreased 5.28%,5.67%,7.16% more respectively in low,medium and high dose of CLA groups than in obesity control groups;ratios of fat to body weight also decreased 52.8%,54.6%,54.6% more respectively in three CLA groups than in obesity control group(P<0.05).Serum triglyceride levels decreased 18.6%,34.8%,49.1% more respectively in three CLA groups than in obesity control group,and significant difference(P<0.05) was found between high dose of CLA group and obesity control group.Fat droplets in liver tissue stained with Oil Red O were decreased as the doses of CLA increased.Conclusion Conjugated linoleic acid can reduce body weight,serum lipid level,and has anti-obesity effect through the influence on liver lipid metabolism.
