【摘要】 目的比较流式反向序列特异性寡核苷酸探针方法(Flow-rSSO)与聚合酶链式反应-序列特异性引物方法(PCR-SSP)在骨髓库人类白细胞抗原(HLA)分型中的应用价值。方法4769例中华骨髓库质控样本中,已采用PCR-SSP方法分型的3509例标本应用FLOW-rSSO方法复检,而1260例已采用FLOW-rSSO方法分型样本则应用PCR-SSP方法复检,结果不一致样本采用PCR-测序分型方法(SBT)进行确认。结果PCR-SSP方法HLA分型错误率(6.84‰)明显高于Flow-rSSO方法(1.59‰),其中PCR-SSP方法中75%的错误结果是由于漏检造成;PCR-SSP方法的模棱两可分型结果比例(2.56‰)明显低于Flow-rSSO方法(16.67‰)。结论PCR-SSP方法和FLOW-rSSO方法均适合应用于中华骨髓库供者的HLA分型,但HLA分型实验室有必要同时建立两种HLA分型方法。更多还原

【Abstract】 Objective To Compare the applications of Flow-rSSO and PCR-SSP methods in bone marrow donors HLA typing.Methods Among the 4769 quality-control samples from Chinese Marrow Donor Program(CMDP),3509 samples genotyped by PCR-SSP were retested by Flow-rSSO method,and other 1260 genotyped by Flow-rSSO were retested by PCR-SSP method.PCR-SBT method were used to confirm the final results which disaccorded in the above two methods.Results The ratio of wrong HLA typing by PCR-SSP method(6.84‰)was significantly higher than that of Flow-rSSO method(1.59‰),and among them,75% wrong HLA typing samples by PCR-SSP were due to missing.Whereas,the ratio of ambiguous typing results by PCR-SSP(2.56‰)is significantly lower than that of Flow-rSSO method(16.67‰).Conclusions The Flow-rSSO and PCR-SSP methods a both conformably applied in HLA typing of bone marrow donor from CMDP.Two different HLA typing methods should be establish for HLA typing detection.更多还原
