紫杉醇衍生物对人喉癌细胞系Hep-2的抑制作用
Inhibition of human laryngeal cancer cells Hep-2 by taxol derivative
【摘要】 目的:通过体外实验评价紫杉醇衍生物GT对人喉癌细胞系Hep-2的抑制作用。方法:应用MTT法测定GT对Hep-2细胞的增殖抑制作用;通过细胞形态学及流式细胞术考察GT对Hep-2细胞诱导凋亡的作用。结果:MTT实验显示GT对Hep-2细胞具有明显的增殖抑制作用;当GT作用细胞后,在光镜下能观察到较为典型的细胞凋亡的形态学变化,如细胞核固缩、染色质凝集成新月型紧贴于核膜周边,核碎裂、染色质断片化、凋亡小体形成等。流式细胞仪分析结果可见G2/M期的细胞比例明显增加,并出现典型的亚二倍体“凋亡小峰”。结论:GT对Hep-2细胞的生长有明显的抑制作用,其效果与紫杉醇基本相同,可使细胞分裂阻滞于G2/M期,并诱导肿瘤细胞凋亡,有进一步研究开发的价值。
【Abstract】 Objective: To assess the inhibitory effect of taxol derivative(GT) on human laryngeal cancer Hep-2 cell line in vitro. Methods: MTT assay,cell morphology and flow cytometry were used to examine GT-induced apoptosis and growth inhibition of human laryngeal cancer Hep-2 cell line.Results: GT could inhibit the proliferation of Hep-2 cells in vitro,which was associated with arrest of G2/M phase and with inducing apoptosis.Chromatin condensation,nuclear fragmentation and apoptic body formation were found.Flow cytometry analysis showed a sub-G1 cell peak.Conclusion:GT could induce apoptosis and block the cell cycle of human laryngeal cancer cells.
