透明颤菌血红蛋白基因在恶臭假单胞菌中的克隆及表达

【摘要】 自然界存在一种革兰氏阴性,专性好氧菌透明颤菌1#(Viteroscilla1#),在微氧条件下,可以诱导合成一种类似于血红蛋白的可溶性物质,即透明颤菌血红蛋白(VitereoscillaHemoglbim,VHb)。受体恶臭假单胞菌6-81(Pseudomonas puti-da6-81)具有降解PTA功能。VHb基因在受体6-81中克隆并获得表达。研究了转化子LEY-9与受体6-81在不同供氧条件下对PTA降解率的差异。在供氧条件下,LEY-9对PTA的降解率比6-81高29.6%;在限氧条件下,LEY-9对PTA的降解率比6-81高39%。转化子LEY-9在限氧条件下,对PTA的降解率比在供氧条件下受体6-81,对PTA的降解率高出20.6个百分点。通过将VHb基因克隆入对PTA具有降解功能的受体菌中,可使其在较低的溶解氧环境中,仍具有较高的降解率。对VHb在污水处理系统中的应用作了初步的探索。

【Abstract】 The Vitreoscilla 1~# was a Gram-negative obligate aerobe.Under the condition of tiny oxygen,it can be induced to synthesize one category of dissoluble material like the hemoglobin,namely the Vitereoscilla Hemoglbim(VHb).The receptor Pseudomonas putida 6-81 had the function of degrading PTA(p-Phthalic acid).The gene of VHb was cloned and expressed in the Pseudomonas putida 6-81.Studied the transformant LEY-9 with the receptor 6-81 under the different conditions that resulted in different PTA degradation rates.Under the condition of providing the oxygen,the rate of PTA degradation caused by the LEY-9 was 29.6% higher;under the condition of limiting the oxygen,the rate of PTA degradation caused by the LEY-9 was 39% higher than 6-81.Under the condition of limiting the oxygen,the rate of PTA degradation caused by the LEY-9 was 20.6% higher than 6-81,under the condition of providing the oxygen.The gene of VHb was cloned in 6-81 cell;it has higher rate of PTA degradation in the lower deliquescence oxygen environment.
