利用双重抑制PCR技术分离鸭微卫星标记
Isolation of duck microsatellite markers using dual-suppression-PCR technique
【摘要】 用双重抑制PCR技术分离10对鸭的特异微卫星引物,应用于北京鸭、樱桃谷鸭、高邮鸭3个品种的基因组PCR扩增。结果表明:10对引物中有7对引物具有多态,3对呈高度多态,4对呈中度多态。7对引物在上述3个鸭品种中分别扩增出23、22、23个等位基因,其中15个等位基因为3个品种所共有;各位点产生4~7个等位基因,平均杂合度为0.494~0.650,平均多态信息含量为0.414~0.586,平均有效等位基因数为2.001~2.870。3个鸭品种的平均杂合度分别为0.578、0.566、0.594,平均多态信息含量分别为0.496、0.480、0.524;平均有效等位基因数分别为2.441、2.340、2.615,与实测平均等位基因数3.286、3.142、3.286较接近。
【Abstract】 A dual-suppression-PCR technique was used to isolate 10 duck special microsatellite primers from duck,then these primers were used to amplify DNA of Peking duck(BJ) and Cherry Valley duck(CV) and Gaoyou duck(GY).The results showed 7 microsatellite loci with polymorphism.There were 3 loci with high polymorphism and 4 loci with middle polymorphism.7 primers could generate 23,22 and 23 alleles in 3 duck breeds,in which there were 15 same alleles.Alleles produced by these loci ranged from 4 to 7 per locus and the mean heterozygosities per locus were calculated as values from 0.494 to 0.650,and the average polymorphism information content(PIC) per locus were from 0.414 to 0.586.On the basic of effective number of alleles(Ne),the statistical values of BJ,CV and GY were 2.441,2.340 and 2.615 close to practical average values 3.286,3.142 and 3.286 respectively.The results indicated that the dual-suppression-PCR technique was of low cost,high efficiency and good polymorphism of microsatellite marker obtained,so it was suitable for studying duck genetic diversity.
